Biological function summary
The enzyme SMPDL3A significantly impacts cellular lipid regulation and membrane dynamics. It does not typically function as part of a complex but stands out in remodeling membrane-bound lipids. SMPDL3A's ability to degrade sphingomyelins affects signaling pathways by altering ceramide levels influencing cellular response processes like apoptosis and proliferation. Therefore its biological role involves influencing membrane composition and cell signaling pathways that respond to various stimuli.
Pathways
This protein significantly participates in the regulation of the lipid metabolism and sphingolipid signaling pathways. It operates alongside other proteins like ceramidase and sphingosine kinase 1. SMPDL3A alters sphingolipid metabolism which influences the MAPK signaling pathway a pathway critical in cell growth and stress responses. By modulating ceramide levels it indirectly affects downstream interactions and protein activities related to these pathways.
일반 정보
기능
Cyclic-nucleotide phosphodiesterase that acts as a negative regulator of innate immunity by mediating degradation of 2',3'-cGAMP, thereby inhibiting the cGAS-STING signaling (PubMed : 37890481). Specifically linearizes 2',3'-cGAMP into 2'5'-bond pGpA and further hydrolyzes pGpA to produce GpA (By similarity). Also has in vitro nucleotide phosphodiesterase activity with nucleoside triphosphates, such as ATP (PubMed : 26792860). Has in vitro activity with p-nitrophenyl-TMP (By similarity). Has lower activity with nucleoside diphosphates, and no activity with nucleoside monophosphates (By similarity). Has in vitro activity with CDP-choline, giving rise to CMP and phosphocholine (By similarity). Has in vitro activity with CDP-ethanolamine (By similarity). Does not have sphingomyelin phosphodiesterase activity (By similarity).
