Rabies Virus Glycoprotein (RVG) also known as G protein plays an essential role in the viral entry process. It has a mass of approximately 59 kDa and expresses on the surface of the rabies virus. The RVG mediates the attachment of the virus to the host cell's receptors facilitating fusion with the host cell membrane. This interaction enables the virus to enter the host cell and initiate infection. The glycoprotein is critical in the determination of host range and tissue tropism due to its binding properties.
Biological function summary

The glycoprotein participates in the fusion process between the viral envelope and host cell membrane. This process requires a conformational change in the glycoprotein structure triggering the fusion of the viral and cellular membranes. RVG forms homotrimers becoming part of a functional complex necessary for viral infectivity. It acts as the major antigenic determinant that elicits an immune response in the host making it a target for neutralizing antibodies.

Pathways

RVG plays a significant role in the viral replication and propagation pathway. Its interaction with cell receptors like the nicotinic acetylcholine receptor and neural cell adhesion molecule (NCAM) facilitates viral attachment and entry into nerve cells. This interaction contributes to the virus's neurotropic characteristics. Glycoprotein interaction with non-classical major histocompatibility complex (MHC) class I proteins can modulate host immune response pathways illustrating its diverse involvement in cell signaling.

Rabies virus causes rabies a fatal neurological condition transmitted through the saliva of infected animals. The RVG's role in viral entry makes it a target for developing anti-rabies therapeutics and vaccines. Glycoprotein interactions with proteins like NCAM could provide insight into therapeutic approaches for controlling viral entry into neurons. Understanding this interaction is important in preventing the spread of rabies and developing effective anti-rabies strategies.

제품 프로토콜

타겟 정보

Attaches the virus to host cellular receptor, inducing endocytosis of the virion by using different host proteins including TFRC, GRM2 and ITGB1 (By similarity). In the endosome, the acidic pH induces conformational changes in the glycoprotein trimer, which trigger fusion between virus and cell membrane. There is convincing in vitro evidence that the muscular form of the nicotinic acetylcholine receptor (nAChR), the neuronal cell adhesion molecule (NCAM), and the p75 neurotrophin receptor (p75NTR) bind glycoprotein and thereby facilitate rabies virus entry into cells (By similarity).
