The severe acute respiratory syndrome coronavirus envelope protein often referred to as the SARS E protein is a small multifunctional protein with a molecular mass of about 9.3 kDa. It is expressed in various locations within the host cell including the Golgi apparatus and endoplasmic reticulum-Golgi intermediate compartment. As a structural component of the virus the E protein plays an important role in virus assembly and release contributing to the formation of the viral envelope.
Biological function summary

The SARS E protein serves specific roles in the virus's life cycle and pathogenicity. It interacts with host cell membranes functioning as a viroporin that forms ion channels. This activity modifies host cell membrane permeability and influences the virus's capacity to assemble and mature properly. Additionally the E protein plays a part in inflammasome activation and is not usually part of a large protein complex but its interactions significantly impact the host cell environment.

Pathways

The SARS E protein engages in processes critical to viral pathogenesis. One major pathway involving the E protein is the ER stress response where it influences host cell stress mechanisms. It also intersects with the NF-kB signaling pathway promoting pro-inflammatory responses. These pathways and their interactions with other viral proteins such as the SARS M protein highlight the functionality of the E protein in the context of the virus's biological lifecycle and its ability to modulate host immune responses.

The SARS E protein is directly linked to the pathogenesis of SARS-CoV contributing to respiratory illness and acute lung injury. The interactions between the E protein and other viral proteins like the N protein exacerbate inflammatory responses a characteristic feature of the disease. Understanding its role could further possibilities in developing therapeutic interventions targeting these pathways or reducing the inflammatory damage associated with SARS-CoV infections.

일반 정보

기능

Plays a central role in virus morphogenesis and assembly. Acts as a viroporin and self-assembles in host membranes forming pentameric protein-lipid pores that allow ion transport. Also plays a role in the induction of apoptosis (By similarity). Activates the host NLRP3 inflammasome, leading to IL-1beta overproduction.

서열 유사성

Belongs to the betacoronaviruses E protein family.
