If this differential immunostaining pattern is specific then I would reclassify this Ab as 'excellent".
In the submitted high power upper image of the high cortical region of kidney, only the smooth muscle cells of the Afferent/efferent arterioles (AAorAE) and a subpopulation of cells ( ?podocytes) in the Glomerulus are clearly positive.
The nuclei of the various collecting tubules ( CT) are either negative or poorly positive.
There is also a slight cytoplasmic positivity in the Collecting tubules (CTs) that is not caused by endogenous biotin as this was blocked prior to immunostaining (adjacent section: omission of primary Ab was unstained, not shown) .
In the lower image of the cortico-medullary region, taken at a lower magnification, the smooth muscle cell nuclei of the Interlobular arteries ( IA) are clearly positive.
It may be that this specimen, kindly donated to me by a Researcher at DEFRA, is relatively inadequately fixed: this may have resulted in degradation of Lamin.
However,if you examine my Human jejunum abreview for this Ab, this also shows a diffferential positivity for this Ab reagent.
So, why would Lamin in some cells be highly labile yet in other cells be very stable? I do not know.
It could be that this Ab reagent is inadequate......not my area of research.
However, in one of my areas of research (neural stem cell markers ) this Ab is also negative for progenitor olfactory bulb cells.
