A pepsin pretreatment is required. Especially useful for immunohistochemical detection of laminin in extracellular matrix or basement membranes in freshly frozen tissue sections of rat, mouse or humans.

Thank you for your enquiry regarding ab2466. The antigen retrieval method of pepsin treatment is specific for this antibody as it was found that staining was significantly better with this step. This method is classically used for formalin fixed paraffin embedded sections but in this instance was found to also help the staining procedure on freshly frozen tissue sections of rat,mouse or humans. If using your other laminin antibodies on frozen sections you do not need to pretreat with pepsin unless advised on the datasheet. Thank you for your enquiry and for your patience. At this time, all the information that we have regarding these two antibodies is located on the online datasheets. Ab6346: MTn-12 (Aufderheide et al. 1988) reacts specifically with mouse tenascin. In supernatants of cultured mouse fibroblasts, the antibody precipitates two polypeptides of ~210 and ~260 kDa. It also recognizes these tenascin chains and the 230 kDa endoglycosidase F-digested fragment in immunoblotting of tissue extracts and fibroblast lysates under reducing conditions (Aufderheide et al. 1988, Weller et al. 1991). In ELISA, it was shown that the antibody reacts specifically against mouse tenascin, but not against fibronectin and collagen type I. Ab2466: This antibody reacts with the P1 fragment of laminin.
