  • ICC/IF image of ab44941 staining human prostate cells.  The cells were fixed with 4% paraformaldehyde, permeabilised with 0.8% Triton X-100, and blocked with 20% serum for 1 hour at 24°C.  The primary anitibody was diluted 1/400 in in 20% horse serum in PBS and incubated for 20 minutes at 4°C.  An Alexa Fluor® 488 conjugated donkey anti-rat was used as the secondary antibody.

    The blue staining is the cell nuclei, the green staining shows the laminin beta 1.

  • ab44941 staining Laminin beta 1 in porcine jejunum tissue by Immunohistochemistry (Frozen sections).
    Tissue was fixed in methanol/acetone, permeabilized using 0.3% Triton, blocked with 1% normal goat serum for 10 minutes at room temperature and then incubated with ab44941 at a 1/800 dilution for 2 hours. The secondary used was an Alexa-Fluor 488 conjugated goat anti-rat IgG (H+L) used at a 1/200 dilution.

1 - You received 4 vials of ab44941. How many of the bottles did you test? I ask incase the issue may be limited to a specific bottle.

2 - You used Low pH antigen retrieval methods for ab44941, but did you also try the High pH antigen retrieval? If so, were the results the same?

When conducting IHC or IF, it is generally not advisable to use antibodies which are raised in the same species as the target sample (e.g. a mouse monoclonal antibody against mouse samples). The secondary antibody may recognize endogenous IgG in the sample tissue as well as the primary antibody, which may lead to non-specific staining and high background.

At Abcam, providing quality service is just as important as providing quality products. Our Abpromise® ensures that you can trust our products to perform as stated on the datasheets, or we will offer a replacement, credit, or refund. While we do not other available information for this in mouse with these applications. These are 100% covered by the Abpromise.

In this instance however you should not have any issues with cross reactivity as the species IgG differences are great enough. However in order to assure limited cross reactivity I would recommend using a pre-absorbed secondary antibody. This means they have been absorbed with other proteins, or serum from variousspecies, to eliminate any antibody that may cross-react. The resulting purified antibody should be very pure and specific and any crossreactivity should be significantly reduced.
