    why is the actual band size different from the predicted?

  • ab30687 staining LAMP1 in fruit fly (Drosophilia melanogaster) S2 cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with paraformaldehyde, permeabilized with 0.25% Triton X-100 in PBS for 10 minutes and blocked with 5% BSA for 1 hour at 25°C. Samples were incubated with primary antibody (1/500 in blocking buffer) for 16 hours at 4°C. A Cy3®-conjugated goat anti-rabbit IgG polyclonal (1/400) was used as the secondary antibody.

    Exposure time: 1 minute


    This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 2% Bovine Serum Albumin before being incubated with ab30687 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution ab133406.
