I appreciate your sharing of the protocol with me. The only reason I think it may be worth trying the 20% gel is that your lower percentage gels may cause the protein to run off your gel since the 16 kDa portion of your ladder is near the end of your gel and the leptin protein can at times show up at 14-15 kDa . Thus, I would try running with the 20% gel, taking care that the 16kDa mark on the ladder does not get close to the end of the gel.

If this does not work, I would like to send you a positive control rat leptin protein (ab73532) free of charge to determine if the antibody is not properly detecting leptin or if your samples simply have levels of leptin that are too low to detect.

Lastly, if the rat leptin protein ab73532 does not yield a band around 16 kDa, I would be happy to send you a free of charge replacement (such as ab7210 that you mentioned).

1. What blocking solution did you use?
2. Did you reduce and denature your samples? We recommend boiling in SDS + beta-mercaptoethanol or DTT for 5 minutes at 95oC prior to loading.
3. What was the percentage of acrylamide in gel? For proteins of 4-40 kDa we recommend 20% acrylamide gel
4. Did you run positive and negative controls?
