  • Function

    Receptor for obesity factor (leptin). On ligand binding, mediates signaling through JAK2/STAT3. Involved in the regulation of fat metabolism and, in a hematopoietic pathway, required for normal lymphopoiesis. May play a role in reproduction. Can also mediate the ERK/FOS signaling pathway.
  • Tissue specificity

    Isoform A is expressed in fetal liver and in hematopoietic tissues and choroid plexus. In adults highest expression in heart, liver, small intestine, prostate and ovary. Low level in lung and kidney. Isoform B is highly expressed in hypothalamus.
  • Sequence similarities

    Belongs to the type I cytokine receptor family. Type 2 subfamily.
    Contains 4 fibronectin type-III domains.
    Contains 1 Ig-like (immunoglobulin-like) domain.
  • Domain

    The cytoplasmic domain may be essential for intracellular signal transduction by activation of JAK tyrosine kinase and STATs.
    The WSXWS motif appears to be necessary for proper protein folding and thereby efficient intracellular transport and cell-surface receptor binding.
    The box 1 motif is required for JAK interaction and/or activation.
  • Post-translational

    On ligand binding, phosphorylated on two conserved C-terminal tyrosine residues (isoform B only) by JAK2. Tyr-986 is required for complete binding and activation of PTPN11, ERK/FOS activation and, for interaction with SOCS3 (By similarity). Phosphorylation on Tyr-1141 is required for STAT3 binding/activation.

We have a number of different antibodies directed against the human Leptin protein. The ideal choices for your experiment would depend on what application you are hoping to use.

We have several antibodies which are able to detect different epitopes from the Leptinprotein such as ab50424 which is a goat polyclonal raised againsta synthetic peptide derived from the residues 829-841 of human Leptin. This antibody has been used in ELISA and western blotting.

The antibodyab20101 (mouse monoclonal)has been raised against the peptide taken from residues 131-145 of human Leptin. This antibody has been used in sandwich ELISA, Western blotting and IHC. The antibody ab20101 was used as an ELISA sandwich pair with the rabbit polyclonal ab7208 which was raised against the full length human Leptin protein. The standard curve was performed using the recombinantly expressed human Leptinprotein (ab51273).

In order to advise which secondary antibody (anti-IgG of host animal) would be suitable, I would need to know which conjugate you would like to use in your assay. It would also be helpful to know the application in which you wish to use the antibodies. If you could provide me with this information I will be able to advise you further. Thank you for your enquiry. I can confirm that the immunizing peptide is in all three reported isoforms (NP_002294.2; NP_001003679.1; NP_001003680.1). NCBI calls them isoforms 1, 2, and 3 resp, but the alternative named isoforms are CRA_b, CRA_c and CRA_d, and also contain the immunizing peptide. Therefore, this product contains the isoform b you are looking for.
