Using overnight incubation and ABC/PAP procedure. Dilutions up to 1/10,000 have been achieved under optimized conditions. 1/200 to 1/4000, using overnight incubation and indirect immunofluorescence procedure. All standard crosslinking agents may be used, but formaldehyde based solutions are recommended.
Frozen sections and Vibratome slices are recommended for tract tracing.
Not tested in other applications.

RelevanceEnkephalin and enkephalin-like peptides have been detected in many animal species. In mammals these opioid pentapeptides are cleaved from a common precursor molecule which is expressed in a number of cell types, including neurons and endocrine cells associated with the sympathetic nervous system where they are often co-stored with catecholamines. They are known to be endogenous ligands for Sopioid receptors and are known to mediate analgesia at the spinal level.

ab22619 (4µg/ml) staining leu enkephalin in human testis using an automated system (DAKO Autostainer Plus). Using this protocol there is cytoplasmic staining of seminal vesicles.
Sections were rehydrated and antigen retrieved with the Dako 3 in 1 AR buffer citrate pH 6.0 in a DAKO PT link. Slides were peroxidase blocked in 3% H2O2 in methanol for 10 mins. They were then blocked with Dako Protein block for 10 minutes (containing casein 0.25% in PBS) then incubated with primary antibody for 20 min and detected with Dako Envision Flex amplification kit for 30 minutes. Colorimetric detection was completed with Diaminobenzidine for 5 minutes. Slides were counterstained with Haematoxylin and coverslipped under DePeX. Please note that, for manual staining, optimization of primary antibody concentration and incubation time is recommended. Signal amplification may be required.
