    Potential transcription factor. May play a role in early mesoderm formation and later in lateral mesoderm differentiation and neurogenesis.

  • ab14554 staining LIM1 in human fetal kidney cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with formaldehyde, permeabilized with 0.5% Triton X-100 in PBS and blocked with 5% BSA for 1 hour at 25°C. Samples were incubated with primary antibody (1/200) for 1 hour at 37°C. An Alexa Fluor® 488-conjugated mouse anti-rabbit IgG polyclonal (1/200) was used as the secondary antibody. Actin filaments stained with phalloidin (red).

  • ab14554 at 1/200 dilution staining mouse postnatal day 5 retina cells (known to express LIM 1) by ICC/IF. The cells were formaldehyde fixed and incubated with the antibody for 72 hours. An Alex-Fluor 488 conjugated donkey anti-rabbit antibody was used as the secondary. The image shows strong horizontal cell process labeling with fainter nuclear label.

Thanks for your reply and I look forward to receiving your image. Regarding the 150 kDa band, I have looked through our records and briefly in the literature and have found no evidence that LIM1 should run that high, so this band is most likely non-specific. Using RIPA buffer will ensure that all of the LIM1 in the nucleus is released, which may increase the strength of the specific band but won't affect the non-specific band. Also, using RIPA (contains SDS) and boiling the sample in reducing and denaturing buffer (like Laemmli) will ensure that the protein is running as a monomer and should be at the expected weight. Please let me know if you have any questions.
