    Answer

    For immunoprecipitation of LRG1 from plasma, we added 6ul of the antibody into 300ul of the plasma lysates (the protein concentration in the lysate is about 3.5mg/ml).

    Plasma lysate preparation
    1. Whole blood with anticoagulant. (Recommend using the fresh whole blood within 6 to 8 hours)
    2. Centrifuge at 5000g for 7min at 2 ˜ 6 ℃;
    3. First layer is the plasma. Gently separate plasma using siphon;
    4. Measure the protein concentration using BCA method.
    5. Adjust the protein concentration of the lysate to 3.5 mg/ml with IP lysis buffer.

    IP lysis buffer:
    20mM Tris-HCl (pH7.5)
    150mM NaCl
    1% Triton
    1mM Na2EDTA
    1mM EGTA
    2.5 mM sodium pyrophosphate
    1mM β-glycerophosphate
    1mM Na3VO4
    1ug/mL Leupeptin
    1mM PMSF
