We would recommend to try using RIPA lysis buffer for the sample preparation if not already tried and see if this achives any improvements of the results.

Furthermore we would recommend to load 40 ug of Protein sample if possible, to prevent overloading the gel.

Considering that you would like to perform IHC on rabbit specimen, I agree that it is advisable to avoid antibodies generated in rabbit. Therefore, although the MMP12 antibody ab56305 shows a rather low sequence similarity across the full-length protein, it might indeed be the most appropriate of our MMP12 antibodies to test, as it is a mouse monoclonal antibody that is guaranteed to work in IHC-P. We have also a chicken polyclonal and a goat polyclonal anti-MMP12 antibody available. As these are not yet tested in IHC though, they might be a better option to test later, in case ab56305 might not work.
