  • ab36362 staining Lysozyme in Human lacrimal gland tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections).

    Tissue was fixed with formalin and blocked with 2.5% BSA; antigen retrieval was by Tris-EDTA buffer. Samples were incubated with primary antibody (1/100 in diluent) for 2 hours at room temperature. A polymer HRP secondary was used and stained with DAB. Counterstaining was by hematoxylin.

Thank you again for your call earlier this week and for letting us know about the trouble with these antibodies.
I have heard from the lab about the protocol used with ab36362, and the sample was prepared in non-reducing buffer, however we have not specifically tested the antibody in reduced conditions so we can not say for sure that non-reduced conditions are required. The antibody was tested at 1:600against 0.5-1 ug/mL of antigen and detected with ECL. It may be a good idea to try one more time using a 1:600 dilution and non-reduced sample (just leave DTT outof the sample buffer), but if you would like to try a different lysozyme antibody I would also be happy to send a free of charge replacement. We also have ab108508, which has been tested with mouse samples in Western blot-

I am currently working to add the blocking peptide of ab66610 to our catalog, and I'll be happy to send a free of charge vial to use in a blocking assay to test the specificity of the band on your WB. The full protocol used to test this antibody in Western blot is attached to this email, and it may be helpful to compare to your current conditions.

I'm waiting to hear back from the lab regarding the protocol used to test ab36362 in Western blot. There is a note on our datasheet stating that it should be done in non-reduced conditions (removing DTT from the sample buffer), but I will forward the full protocol to you as soon as I have it.

We do fully guarantee these antibodies to work as stated on the datasheets, so if the results are not satisfactory we can send replacement antibodies or alternatively issue credits or refunds. Please let me know if you would prefer any of these options at any time.

As you have been using the antibody successfully in Western blotting it seems that the antibody is functioning properly, but it may be that some optimisation of the IHC protocol may be required.

In order to understand the problems encountered more fully and hopefully be able to provide protocol suggestions I am attachinga questionnaire to gather further information.OnceI havereceived the completed questionnaire,I will look at the protocol and see if there are any suggestionsI can make that may improve the results. It wouldbe very useful if you could include some images of the staining obtained, as well asan image of the blot producedwhen the antibody wasused in Western blotting.

Merci pour votre patience. Malheureusement je n'ai pas encore pu conclure pour savoir quel anticorps sera le meilleur pour réagir contre la protéine de poulet et humaine en même temps. Deux de nos anticorps sont faites contre un peptide (ab108508, ab91653) et j'attends la réponse du laboratoire pour connaître l'alignement de ce peptide avec la séquence de poulet. Autrement, nos anticorps reconnaissent soit la protéine de poulet (ab34799, ab20596 ,ab34620) soit la protéine humaine (ab2408, ab76784, ab2408, ab9011, ab93324, ab36364, ab942, ab74666). Le ab74666 est testé contre plusieurs espèce, comme Hu, Bb, Cat, Dog, Mk, Pig, mais pas le poulet. L'alignement entre la protéine de poulet et la protéine humaine n'est pas si mal. http://www.uniprot.org/align/201109192CDUKL4DKS Je pense qu'il serait probable que aussi un anticorps comme le ab74666 ou ab34799, pourront reconnaitre la protéine de poulet.
