    Ligand-specific transporter trafficking between intracellular organelles (TGN) and the plasma membrane. Plays a role in autocrine regulation of cell growth mediated by growth regulators containing cell surface retention sequence binding (CRS). May act as a hyaluronan (HA) transporter, either mediating its uptake for catabolism within lymphatic endothelial cells themselves, or its transport into the lumen of afferent lymphatic vessels for subsequent re-uptake and degradation in lymph nodes.

    ab36993's immunogen is a recombinant full length protein produced in insect cells, corresponding to amino acids 24-232 of Human LYVE1 and is fused to a C-terminal His-tag (6xHis). This antibody is therefore not specifically directed towards the C-terminal of LYVE-1.
    However, we have Anti-LYVE1 antibody (ab33682) which is guaranteed to work in IHC-P and it's immunogen is a synthetic peptide conjugated to KLH derived from within residues 300 to the C-terminus of Mouse LYVE1.

    Thank you for your replay.

    That will be really helpful if you will send us the replacement (ab10278), and am really hoping it will give us beautiful staining results.

    Always I would like to get the good antibody for IF staining. Since mouse LYVE1 antibody from your company works very well, so I ordered ab36993.

    But it didn’t work for paraffin slide in IF staining for any condition.

    (It may work for frozen, but I couldn’t get the human patient frozen sample.)

    So I would like to try another human LYVE1 antibody from your company.





    The key factors you need to know is here.

    What human tissue is tested?---human H&N patient paraffin slides

    Duration of retrieval?----10 minutes in pressure cooker

    Blocking conditions (x% BSA/Serum in TSB/PBS for 1h/ overnight)?----10% Serum in 0.01% tween TBS

    Primary antibody incubation conditions?-----4 degree over night



    Hope this information is enough. Please let me know if you need to know more about my staining condition.

    I tried to make sure if any condition can work with this antibody (LYVE1 ab36993), but still hard to get clear staining results.

    Sorry, it was taking time for making sure, so my replay was too late.

    The problem for IF, I couldn't get staining with paraffin slides, so I didn’t put the picture, but about IHC, I attached the picture.

    I have checked our records and saw that we are still waiting for you to return our questionnaire to us.     Question

    I have a question about the primary antibody from your company.




    I bought the primary antibody for human LYVE1 ab36993, but I couldn’t get staining with human sample.

    I did IF and IHC both, and also tried to use pH6.0, pH7.0 citric acid, and pH8 EDTA for antigen retrieval.

    Is there any way to get the good human LYVE1 antibody instead of this antibody without paying extra money?

    I am sorry to hear you are experiencing difficulties with one of our products. We take product complaints very seriously, and investigate every product that we feel may not be performing correctly.

    I am attaching our questionnaire so that we can gather further information regarding the samples tested and the protocol used. Once we receive the completed questionnaire, we will look at the protocol and see if there are any suggestions we can make that may improve the results. This information will also allow us to investigate this case internally and initiate additional testing where necessary.
