WB Use at an assay dependent concentration. To detect hM-CSF by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hM-CSF is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.
ELISA Use at an assay dependent concentration. To detect hM-CSF by direct ELISA (using 100µl/well antibody solution) a concentration of at least 0.5µg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 - 0.4 ng/well of recombinant hM-CSF.
Neutralising Use at an assay dependent concentration. To yield one-half maximal inhibition [ND50] of the biological activity of hM-CSF (7.0 ng/ml), a concentration of 0.02 - 0.03 µg/ml of this antibody is required.
Sandwich ELISA Use a concentration of 0.1 µg/ml.

For sandwich ELISA, use this antibody as Detection at 0.1µg/ml with ab18645 as Capture.

IHC-P Use a concentration of 5 µg/ml.

    Granulocyte/macrophage colony-stimulating factors are cytokines that act in hematopoiesis by controlling the production, differentiation, and function of 2 related white cell populations of the blood, the granulocytes and the monocytes-macrophages. CSF-1 induces cells of the monocyte/macrophage lineage. It plays a role in immunological defenses, bone metabolism, lipoproteins clearance, fertility and pregnancy.

    modifications
    Glycosylation and proteolytic cleavage yield different soluble forms. A high molecular weight soluble form is a proteoglycan containing chondroitin sulfate.
    Isoform 1 is N- and O-glycosylated. Isoform 3 is N-glycosylated.

  • ab9693 staining MCSF in human cervical squamous cell carcinoma section by Immunohistochemistry (Formalin/PFA fixed paraffin-embedded sections). Tissue underwent heat mediated antigen retrieval in sodium citrate buffer (pH 6.0). The primary antibody was used at 5 ug/ml and incubated with sample at 4°C overnight. A HRP-labeled polymer detection system was used with a DAB chromogen.

We currently only have one product which is guaranteed to work in mouse, however, as we have spoken about, this product is currently unavailable in the US.

We do have two other products which have been shown to work in neutralizing assays. These are the rabbit polyclonal antibody ab9693 (https://www.abcam.com/MCSF-antibody-ab9693.html) and a mouse monoclonal, ab89571 (https://www.abcam.com/MCSF-antibody-ab9693.html).

I was able to aquire the immunogen sequence used for Ab9693. This sequence had an 80% identity and 91% positive against the related mouse protein. The polyclonal and would recognize multiple epitope sites within this region as compared to a monoclonal which would only recognize one site which may not be present in mouse protein. As such I would recommend ab9693 for your studies.
