Application Abreviews Notes
WB Use at an assay dependent dilution. To detect hMIP-1-alpha by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hMIP-1-alpha is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.
ELISA Use at an assay dependent dilution. To detect hMIP-1-alpha by direct ELISA (using 100µl/well antibody solution) a concentration of at least 0.5µg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 - 0.4 ng/well of recombinant hMIP-1-alpha.
Neutralising Use at an assay dependent dilution.
Sandwich ELISA Use at an assay dependent dilution. Can be used as Detection antibody.

    Monokine with inflammatory and chemokinetic properties. Binds to CCR1, CCR4 and CCR5. One of the major HIV-suppressive factors produced by CD8+ T-cells. Recombinant MIP-1-alpha induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV).

Thank you for your enquiry. I am sorry to hear that you are experiencing difficulties with this product ab9673 in IHC. Often it is possible to make suggestions that help resolve problems. We will happily offer technical support but please bear in mind that this product has not been tested in IHC application and therefore, our Abpromise is not applicable. I have looked through the protocols you used and have a few questions and suggestions that might help you resolve the problem. I noticed that the primary incubation time was 24hr to 72hr, which maybe too long. Can you please try incubating overnight (not more than 18 hours) at 4oC, if you have not already done so. Increasing the dilution to 1:1000 ~ 1:5000, and increasing the washing duration will also help eliminate high background. You can try washing for 3 x 15min. Can I confirm that in your negative controls, omission of the primary antibody no longer cause high background? If so, then the problem lies with the primary antibody. It could be binding non-specifically to your sample. If the above suggestions did not prove to help you get better results, then I am afraid that this product is indeed not suitable for IHC application.
