Target

  • Function
Membrane glycoproteins implicated in the pathologic deposition of cholesterol in arterial walls during atherogenesis. Two types of receptor subunits exist. These receptors mediate the endocytosis of a diverse group of macromolecules, including modified low density lipoproteins (LDL). Isoform III does not internalize actetylated LDL.
  • Tissue specificity
Isoform I, isoform II and isoform III are expressed in monocyte-derived macrophages.
  • Sequence similarities
    Contains 1 collagen-like domain.
    Contains 1 SRCR domain.
  • Cellular localization
    Membrane.

Question
Answer

Thank you for your enquiry. We are sorry to inform you that there is no protein concentration for polyclonal antibodies that are not affinity purified. This antibody is provided as whole serum and the IgG concentration has not been determined.

Answer

All the information we have on species cross reactivity is specified on the datasheet. To our knowledge, cross reactivity with mouse has not yet been tested. Should you decide to go ahead and purchase this product, please let us know how you get on and in return we will forward a reward of your choice, typically an Amazon gift voucher. We do not currently have a mouse-class A macrophage scavenger receptor antibody. Question
Answer

We do not have any reports on the blocking activity of this antibody. The active domain on the macrophage scavenger receptor 1 is described as a simple domain consisting of a cluster of lysine residues. However, we do not know whether this relates to the amino acid sequence the antibody has been raised against. Therefore, the blocking activity of this polyclonal is not determined.

Answer

We recommend a working dilution of 1/200 which, with a serum, corresponds to roughly 0.25 to 5ug/ml per slide. You may also wish to try a lower concentration. Alternatively, you may wish to increase a little the concentration of the blocking serum(5-10%), maybe diluting the primary in this, or decreasing incubation times - all these could improve results. A positive control could be Alzheimer's disease brain sections.

Answer

Details concerning recommended dilutions for this product are indicated on the datasheet for the applications for which it has been tested. However, this antibody is supplied as a polyclonal serum and from experience we can estimate the concentration to be around 0.5-1.0mg/ml, therefore 10ug/ml range could be determined from this. Most other applications for which this antibody has not been tested such as cryostat sections are likely to use a similar concentration as for paraffin and therefore I would suggest this as a starting dilution.

Question
Answer

This antibody is supplied as whole goat antiserum; the concentration of specific antibody in the serum has not been determined.

Answer

For this antibody we recommend an approximate working dilution of 10ug/ml. However, we do suggest using a number of dilutions around this to establish the optimum concentration for individual systems.

Question
Answer

Given the immunogen used, this polyclonal antibody will most probably recognise all isotypes of the Macrophage Scavenger Receptor Protein 1.
