Thank you for the reply...The reaction buffer is to activate the sensor, so after the sensor gets activated, can i lower the pH by adding another buffer to about 7. Based on the protocol you have for this product, first is to prepare the reaction mixture and incubate at RT for 30 min (that is the step to activate the sensor, right), then prepare the maleimide assay mixture (what is the pH for this one) and finally add the analyst into the mixture....My concern is once my subject adds to the assay mixture, my subject will precipitate out. So i want to know if i make the buffer to be neutral after the sensor activation, will it affect the accuracy of this assay.

I have confirmed with the laboratory that regarding the reaction buffer, the pH of this cannot be altered.
You are correct in that the first step is to prepare the reaction mixture and incubate for 30 minutes. Then you prepare the maleimide assay mixture which is pH 7 and add the analyst into the mixture. At this point, as long as your sample's pH is neutral, it will not affect the final pH (7-7.5) and the reaction will be ok.
