Inquiry: I read through the protocol and came to 2 questions. 1. Instead of preparing reagents for a 96 well plate which, can I reduce the amount and measure 6-10 samples at a time? Would it affect sensitivity? I realise that the reagents only last for 1-2 hrs after being prepared. I don't have that many samples to test at a time. 2. Can this kit assay maleimide groups modified onto lipid nanoparticles, which are turbid samples?

I can confirm that the Maleimide Quantification Assay Kit (Fluorometric) (ab112141) can be used for measuring 6-8 samples at a time instead of 96. Please follow the instructions below:
- Aliquot 500X MalemGreen DMSO stock solution (step 1), and store at -20C. Use enough for 6-8 samples for step 2 (such as at the portion of 1 uL of 500X MalemGreen stock solution (from Step 1) into 25 uL Reaction Buffer) and the rest of the steps for the reagent preparations.
The Maleimide Quantification Assay Kit (Fluorometric) (ab112141) should detect maleimide groups modified onto lipid nanoparticles. However, please be aware that the rate of maleimide sensor bound to maleimide groups on nanoparticles might be different from standard. Therefore, we suggest to use your own maleimide-nanoparticles as standard.
