    Malondialdehyde (MDA) is a natural product formed in all mammalian cells as a product of lipid peroxidation. MDA is a highly reactive three carbon dialdehyde produced as a byproduct of polyunsaturated fatty acid peroxidation and arachidonic acid metabolism. MDA readily combines with several functional groups on molecules including proteins, lipoproteins, and DNA. It reacts with DNA to form adducts to deoxyguanosine and deoxyadenosine. The major adduct to DNA is a pyrimidopurinone called M1G which appears to be a major endogenous DNA adduct in human beings that may contribute significantly to cancer linked to lifestyle and dietary factors. MDA modified proteins may show altered physico chemical behavior and antigenicity. MDA is toxic and has been implicated in aging mutagenesis, carcinogenesis, diabetic nephropathy and radiation damage. Increased expression of MDA has been reported in the brains of Alzheimer's patients. Antibodies to MDA will help to visualize the MDA adducts.

  • ab6463 staining Malondialdehyde in human rectal cancer tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections).
    Tissue was fixed with formaldehyde and a heat mediated antigen retrieval step was performed using citrate buffer pH 6.0. Samples were then permeabilized, blocked using 5% serum for 1 hour at 4°C then incubated with ab6463 at a 1/500 dilution for 24 hours at 4°C. The secondary used was a biotin conjugated goat anti-rabbit polyclonal, used at a 1/500 dilution.

  • Immunohistochemical analysis of rat hippocampus tissue, sub-regions CA1, CA3 and DG, staining Malondialdehyde with ab6463. Rats were treated with saline (control) or multiple doses of methamphetatime.

    Sections were fixed in paraformaldehyde and blocked with 10% normal horse serum for 2 hours at room temperature. Samples were incubated with primary antibody (1/200) overnight at 4°C. Staining was detected using DAB.

Unfortunately it's difficult to say exactly what type of cell is staining for malondialdehyde. Since malondialdehyde is an organic compound, it may not be cell-specific. I found some references which may point to location in the mucosa and I've included a few other references below:

Ab6463 has been tested in and is guaranteed to work in WB, IHC-Fr, ELISA and IHC-P. When using in IHC-P we recommend heat mediated antigen retrieval using citrate buffer pH 6.0, generally heating between 10-20 minutes is adequate. I have included Abcam antigen retrieval protocol for your further review. We recommend use of a starting dilution within the range 1/1000-1/5000 however you may find that the optimal dilution in your experiments is outside of this range and need to determine this in the lab. The counter stains and mounting media list should not interfere with the efficacy of the antibody and kit.

Atendiendo al protocolo seguido, me gustaría hacer un par de sugerencias para tratar de optimizar el resultado.
- Es muy posible que el motivo por el que no se observa ninguna tinción se deba a un exceso en las condiciones de bloqueo del tejido. Yo te propondría bloquear usando sólo 5%BSA o solo 5% de suero NGS, pero combinar ambos y usar esta solución para diluir los anticuerpos puede estar impidiendo la tinción del anticuerpo.
Mi recomendación es bloquear solo con BSA o suero y diluir los anticuerpos en TBST.
- Siempre recomendamos optimizar el proceso de recuperación antigénica. Es importante que los buffers estén al pH adecuado, y para optimizar este paso recomendaría probar distintos tiempos de recuperación. Nosotros normalmente llevamos a cabo el proceso de recuperación de la proteína a diferentes tiempos (5, 10, 15, 20, 25 y 30 minutos) con diferentes buffers. Te aconsejo usar tris EDTA, que suele dar buenos resultados.
Quizás al implementar estos cambios se obtengan mejores resultados. Te animaría a probarlo, y en caso de tener cualquier problema contactarme de nuevo.
Si prefieres recibir directamente la nota de crédito, sin tratar de optimizar el protocolo, no habría ningún problema en hacerlo. Simplemente indícamelo para que te envíe la nota de abono.
