FunctionBinds CpG islands in promoters where the DNA is methylated at position 5 of cytosine within CpG dinucleotides. Binds hemimethylated DNA as well. Recruits histone deacetylases and DNA methyltransferases. Acts as transcriptional repressor and plays a role in gene silencing. Functions as a scaffold protein, targeting GATAD2A and GATAD2B to chromatin to promote repression. May enhance the activation of some unmethylated cAMP-responsive promoters.

Formalin-fixed, paraffin-embedded human prostate carcinoma tissue stained for MBD2 using ab224827 at 1/200 dilution in immunohistochemical analysis. Detection: DAB staining.
MBD2 was immunoprecipitated from HeLa (human epithelial cell line from cervix adenocarcinoma) whole cell lysate with ab224827 at 3 µg/mg lysate. Western blot was performed from the immunoprecipitate using ab224827 at 1 µg/ml.
Lane 1: ab224827 IP in HeLa whole cell lysate.
Lane 2: Control IgG IP in HeLa whole cell lysate.
Detection: Chemiluminescence with exposure time of 3 seconds.
