My colleague is currently out of the office.
The source of the antibodyconfirmed that this antibody should show a band at apparent MW of 45kDa provided the samples are processed correctly. They have seen dimerization of certain GPCR proteins upon repeated freezing and thawing cycles, this was apparent in Dopamine D2 receptor which dimerizes and appear as higher MW bands upon heating and thawing.
Theywould suggest not to heat the protein sample in SDS-PAGE sample buffer rather incubate at 37ºC for 30-45min and then apply on the gel.
The size of this protein varies by several kDa in different cell lines probably due to post-translational modifications.
