    • Function

      Acts as component of the MCM2-7 complex (MCM complex) which is the putative replicative helicase essential for 'once per cell cycle' DNA replication initiation and elongation in eukaryotic cells. The active ATPase sites in the MCM2-7 ring are formed through the interaction surfaces of two neighboring subunits such that a critical structure of a conserved arginine finger motif is provided in trans relative to the ATP-binding site of the Walker A box of the adjacent subunit. The six ATPase active sites, however, are likely to contribute differentially to the complex helicase activity. Required for the entry in S phase and for cell division.

      Phosphorylated on Ser-108 by ATR in proliferating cells. Ser-108 proliferation is increased by genotoxic agents. Ser-40 is mediated by the CDC7-DBF4 and CDC7-DBF4B complexes, while Ser-53 phosphorylation is only mediated by the CDC7-DBF4 complex. Phosphorylation by the CDC7-DBF4 complex during G1/S phase is required for the initiation of DNA replication.

      Question

      A customer needs an ab to human MCM2 for IHC and IF. The closest I found is ab6153, but it was not tested for IF. Would you expect it to be applicable for IF? (We know that the abpromise will not be valid for IF).

      Answer

      The antibody has good chances of working in other types of immunohistochemistry other than those already tested as it has the ability to recognise the native form of the protein. If the customer means IF as in ICC/IF I would recommend to try fixing the cells in PFA 5-10min. If the customer means IF as in IHC-P or IHC-Fr with an IF detection method I would recommend to follow our recommendation for those methods but use an IF amplification kit (i.e either a biotin secondary + extra-avidin/FITC or TRITC method or more sensitive methods which come from Kits eg the TSA kit).       Answer

      All the information we have for this product for use in IHC is on the datasheet, including recommended dilutions and advice for antigen retrieval. I also suggest that you click through to the review submitted by a researcher who successfully used this antibody in IHC on human lung cancer paraffin embedded tissue. In terms of the "success rate" this will depend on a number variables, including the species that you are studying. Species cross-reactivity for this product has not been tested. However, as discussed this antibody works very well in IHC on human tissue.
