  • Application notes
    Neutralization:To yield one-half maximal inhibition [ND50] of the biological activity of mMCP-5 (100.00 ng/ml), a concentration of 2.0 - 3.0 µg/ml of this antibody is required.

    ELISA:To detect mMCP-5 by direct ELISA (using 100µl/well antibody solution) a concentration of at least 0.5µg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 - 0.4 ng/well of recombinant mMCP-5.

    Western Blot:To detect mMCP-5 by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant mMCP-5 is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

    Thank you for contacting Abcam. I am sorry that you are having trouble with ab9737. From what I have read about MCP5, it does have a predicted molecular weight of ~12kDa. I have a few questions/suggestions about the protocol that you are using: 1 - Does your transfer buffer contain SDS? I ask as SDS in the transfer buffer does hinder small proteins significantly from binding to the membrane. Also it is recommended to keep the methanol concentration to below 20%. 2 - How long do you boil your samples for? It could be that the MCP5, is complexed with other proteins or in a dimer with itself and so by increasing to a 10 minutes boiling, you would break apart these complexes. 3 - I would suggest increasing the block and incubation to 5% BSA as that may help reduce some of the non-specific bands. If you have already tried the above suggestions or if they fail to work then ab9737 is covered under our Abpromise and so I would be happy to either send you a replacement or a refund.
