      MED13 is a member of the mediator complex that can act as both a positive and negative regulator of transcription. The mediator complex interacts with RNA pol II and its associated proteins, the general transcription factors. Mediator proteins serve as a bridge connecting the activator and basal transcription machinery.

    This Fast-Track antibody is not yet fully characterised. These images represent inconclusive preliminary data.

    • ICC/IF image of ab49468 stained HeLa cells. The cells were 4% PFA fixed (10 min), permabilised in 0.1% PBS-Tween (20 min) and incubated with the antibody (ab49468, 5µg/ml) for 1h at room temperature. 1%BSA / 10% normal goat serum / 0.3M glycine was used to block non-specific protein-protein interactions. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red). DAPI was used to stain the cell nuclei (blue). This antibody also gave a positive IF result in Hek, Hep and MCF7 cells fixed in PFA and HeLa, Hek293, HepG2 and MCF7 fixed in 100% methanol, all at 5ug/ml. However, this Fast-Track antibody is not yet fully characterised. This image represents inconclusive preliminary data.

    Thank you for your enquiry. ab49468 is considered a "Fast track" antibody. Fast track antibodies have only minimal characterization and as such are offered at a reduced price compared to our typical primary antibodies. ab49468 has been tested for staining HeLa cells, and all the protocol details are found in the caption below the figure on our online datasheet. I believe you might use goat serum for blocking in this case? If so, other blocking solutions can be used. PBS with 1% BSA is a common blocking solution used for ICC and can be used with ab494638.
