      Acts as a scaffold for the formation of a ternary MAP3K2/MAP3K3-MAP3K5-MAPK7 signaling complex. Activation of this pathway appear to play a critical role in protecting cells from stress-induced apopotosis, neuronal survival and cardiac development and angiogenesis.

      Binds MAP3K2/MAP3K3 and MAPK7 via non-overlapping residues of the OPR domain. This domain also mediates interactions with SQSTM1 and PARD6A.

      Yersinia yopJ may acetylate Ser/Thr residues, preventing phosphorylation and activation, thus blocking the MAPK signaling pathway.

    • Anti-MEK5 antibody [EP648Y] (ab45146) at 1/1000 dilution (purified) + HeLa (Human cervix adenocarcinoma epithelial cell) whole cell lysates at 20 µg

      Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/20000 dilution

      Predicted band size: 49 kDa
      Observed band size: 49 kDa

      Blocking and diluting buffer: 5% NFDM/TBST

    • Immunocytochemistry/ Immunofluorescence analysis of JAR (Human placenta choriocarcinoma epithelial cell) cells labeling MEK5 with Purified ab45146 at 1:250 dilution (8.1μg/ml). Cells were fixed in 4% Paraformaldehyde and permeabilized with 0.1% tritonX-100. Cells were counterstained with Ab195889 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) 1:200 (2.5 μg/ml). ab150077 Goat anti rabbit IgG(Alexa Fluor® 488) was used as the secondary antibody at 1:1000 dilution. DAPI nuclear counterstain. PBS instead of the primary antibody was used as the secondary antibody only control.

    • Flow Cytometry analysis of HeLa (Human cervix adenocarcinoma epithelial cell) cells labeling MEK5 with purified ab45146 at 1:200 dilution (10 µg/ml) (red). Cells were fixed with 4% paraformaldehyde and permeabilized with 90% methanol. A Goat anti rabbit IgG (Alexa Fluor® 488) secondary antibody was used at 1:2000 dilution. Isotype control - Rabbit monoclonal IgG (Black). Unlabeled control - Cell without incubation with primary antibody and secondary antibody (Blue).

    • Immunofluorescent staining of HeLa (Human epithelial cell line from cervix adenocarcinoma) cells using unpurified ab45146.

    • Anti-MEK5 antibody [EP648Y] (ab45146) at 1/10000 dilution (unpurified) + HeLa (Human epithelial cell line from cervix adenocarcinoma) cell lysate

      Observed band size: 49 kDa

    • Unpurified ab45146 at 1/100 staining HeLa (Human epithelial cell line from cervix adenocarcinoma) cells by ICC/IF.

      The cells were paraformaldehyde fixed and blocked with goat serum prior to incubation with the antibody for 45 minutes. An Alexa Fluor® 488 conjugated goat polyclonal antibody was used as the secondary. This antibody detects MEK5 in HeLa cells as a cytoplasmic protein.

    Thank you for contacting us. I am sorry you are experiencing difficulty with this antibody. Typically when there are black dots on the blot, the antibody is binding with the blocking agent and it is necessary to filter the blocking agent. Was the same blocking agent used with these other antibodies? We have had no reports of contamination for this lot but will be happy to replace the product it is not working well.

    I can confirm that ab45146 antibody is sold as tissue culture supernatant. Unpurified antibodies, such as those sold as whole antiserum, ascites or tissue culture supernatant will not have a concentration stated on the datasheet. Antibody concentration is usually determined by protein assay; serum / ascites / tissue culture supernatant will contain a lot of other proteins, which means the antibody quantification would not be accurate.

    I can confirm that for tissue culture supernatant, concentration of antibody is known to very between 1 - 3 mg/ml
