Target

  • Relevance

It has been reported that the pathogenesis of melanoma proceeds through multiple stages, ranging from benign proliferation of melanocytic cells to acquisition of the capacity to invade tissues and metastasise. Cell surface antigens that are expressed by melanocytes are not expressed by melanoma cell lines.

    Question

Thanks very much for your email and protocol. You see, I talked about your Mel.2 Ab with my boss for our Dynabeads experiment. We are really interested in this Ab, since they are targeting the outside of the melanocytes. However, there is no any reference about relevant study at this point, it would be reasonable to send us a little bit Ab as a sample for letting us to try it on. If our experiment is going well, It could be served as your future reference.

    Question

Is it known if the epitope this antibody recognizes is extracellular? We are hoping to isolate a population of live melanocytes. We will first validate the antibody by ICC. Please send the ICC protocol.

    Answer

We have not performed any epitope mapping with this antibody. While it is known that the antibody is localized in or on the cell plasma membrane, but we do not know the specific region of binding. This antibody has been successfully tested for use in ELISA using fresh melanoma cells and in IHC-frozen at 8um thin sections. Successful fluorescence staining of melanoma cells on glass has been observed after being, washed, fixed for 30 seconds using ice cold acetone, and air dried. This is the extent of the protocol information we have available to share from localization experiments. I have included our ICC protocol with this email.
