    Mesenchymal Stromal Cell Marker (CD44, CD90, Sca-1, CD45, CD29) Antibody Panel ab93759 contains mouse reactive antibodies for key mesenchymal stromal cell markers, including a CD44 rat monoclonal, CD90 rat monoclonal, Sca-1 rat monoclonal, CD45 rat monoclonal and CD29 rat monoclonal antibody.

    The Mouse Multipotent Mesenchymal Stromal Cell Marker Antibody Panel is designed for the characterization of cultured or isolated mouse multipotent mesenchymal stromal cells. The panel contains a group of antibodies for the positive (CD44, CD29, Sca-1, CD90) and negative (CD45) selection of this lineage. Multipotent mesenchymal stromal cells (MSCs) represent a heterogeneous subset of stromal cells. They can be bone marrow-derived or isolated from many other different adult tissues including periosteum, trabecular bone, adipose tissue, and synovium. They exhibit the potential to give rise to cells of diverse lineages including adipocytes, chondrocytes and osteocytes.

  • IHC image of ab25340 staining CD44 on Mouse Tissue sections (T-cell lymphoma). The cells were 4% PFA fixed (10 min) and then incubated in Perkin-Elmer TNB Blocking Reagent at 0.5% as blocking agent for 30 mins at room temperature to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab25340, 1/250 dilution) overnight at +4°C. The secondary antibody was ab7097.
  • ICC/IF image of ab51317 staining Sca-1 in mouse embryonic stem cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab51317, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rat IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue).

Most of the antibodies in this panel has been tested for IHC-P, but not ab3105 against CD90/Thy1. If you wish to try this one for IHC-P you can use the dilution for IHC-Fr, 1-10 ug/ml. You can link to the individual datasheet from the "components" section where it lists each antibody and the applications and look at the specific information for each antibody.

3 of the 4 antibodies in the kit have are guaranteed to work in flow cytometry on mouse tissue. However, the component antibody ab51317 (anti-Sca1) is untested in this application and so we cannot guarantee that it will work for you in this application.

What I could do, is issue you a testing discount code, to say that you will purchase the Stromal Cell Marker panel (ab93759)and will use it in flow cytometry, but specifically submit feedback on howab51317 works in flow cytometry, in the form of an Abreview. This panel was compiled because all antibodies can be used in IHC.

I agree that all antibodies apart from ab51317 (Anti-Sca1 / Ly6A/E antibody [E13 161-7])are tested and guaranteed for flow cytometry and that this application would be a good choice for characterization experiments.

I am sorry I will not be able to extent our guaranteed to all the antibodies for flow cytometry.

I have checked the antibodies about suitability with flow, and except the ab51317 all are tested for this application.

As always, if ab51317 should have been unsuitable, we would have mentioned this on the datasheet, therefore, the antibody simply hasn't been tested in Flow. Unfortunately, we cannot offer our testing discount on panels.

Our Mesenchymal Stromal Cell Marker Panels (human: ab93758, mouse: ab93759) consist of five un-conjugated primary antibodies that can be used to characterise mesenchymal stromal cells.

The antibodies are not conjugated and therefore require a secondary antibody with a fluorochrome.

You can choose these fluorochromes according to the FACScan system. Please contact the originator of the FACScan to ask which fluorochromes are suitable/possible to use.
