    why is the actual band size different from the predicted?
    Additional bands at: 45 kDa. We are unsure as to the identity of these extra bands.


    Exposure time: 12 minutes


    The band observed at 99 kDa could potentially be a cleaved form of Metabotropic Glutamate Receptor 2 due to the presence of an 18 amino acid signal peptide. Metabotropic Glutamate Receptor 2 also contains a number of potential glycosylation sites (SwissProt) which may explain the fuzzy banding pattern observed.
  • Immunohistochemical analysis of mouse cortical neurons, staining Metabotropic Glutamate Receptor 2 with ab15672. Donkey anti-mouse 6-nm colloidal gold particles were used to detect staining. Images were visualized by electron microscopy.

I am glad to read that most of the antibodies you ordered are giving good results. For the two that are not, could you please send some details of the samples and protocols and results? In particular, we would like to know the species, the time in PFA, the antigen retrieval procedure if applied, and the amount of antibody and incubation time. We do no require images but a description of the stain or lack of stain will be useful for our records.

In general, we always ask for this information when we receive a complaint, in the hope that we can resolve the issue with a protocol modification. I am assuming you have a good protocol, samples, and reagents including the secondary antibody, and that you are confident the proteins are expressed in this tissue.

Please note that our guarantee applies to the applications and species listed on the datasheets.

In order to process the replacements I will need the order numbers for the antibodies that failed. Can you please send either the PO number of the Abcam reference number for the orders of each of these antibodies, ab15672 and ab12093, when you send the sample/protocol/results information?
