|WB||Use a concentration of 1 µg/ml. Detects a band of approximately 99 kDa (predicted molecular weight: 104 kDa). See reference Neki et al, Neurosci Letts.|
|IHC-FoFr||Use at an assay dependent concentration. PubMed: 23129762|
FunctionReceptor for glutamate. The activity of this receptor is mediated by a G-protein that inhibits adenylate cyclase activity. May mediate suppression of neurotransmission or may be involved in synaptogenesis or synaptic stabilization.
Tissue specificityWidely expressed in different regions of the adult brain as well as in fetal brain.
Sequence similaritiesBelongs to the G-protein coupled receptor 3 family.
Cellular localizationCell membrane.

Anti-Metabotropic Glutamate Receptor 2/MGLUR2 antibody [mG2Na-s] (ab15672) at 1 µg/ml + Brain (Rat) Tissue Lysate at 10 µg
Goat Anti-Mouse IgG H&L (HRP) preadsorbed (ab97040) at 1/5000 dilution
Developed using the ECL technique.
Performed under reducing conditions.
Predicted band size: 104 kDa
Observed band size: 99 kDa why is the actual band size different from the predicted?
Additional bands at: 45 kDa. We are unsure as to the identity of these extra bands.
Exposure time: 12 minutes
The band observed at 99 kDa could potentially be a cleaved form of Metabotropic Glutamate Receptor 2/MGLUR2 due to the presence of an 18 amino acid signal peptide. Metabotropic Glutamate Receptor 2/MGLUR2 also contains a number of potential glycosylation sites (SwissProt) which may explain the fuzzy banding pattern observed.
Immunohistochemical analysis of mouse cortical neurons, staining Metabotropic Glutamate Receptor 2/MGLUR2 with ab15672. Donkey anti-mouse 6-nm colloidal gold particles were used to detect staining. Images were visualized by electron microscopy.
