    G-protein coupled receptor for glutamate. Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and modulates the activity of down-stream effectors. Signaling activates a phosphatidylinositol-calcium second messenger system and generates a calcium-activated chloride current. Plays an important role in the regulation of synaptic plasticity and the modulation of the neural network activity.

  • Ab27190, at a dilution of 1/50, staining mGluR5 in Paraffin embedded formalin fixed human brain tissue section.
  • ICC/IF image of ab27190 stained PC12 cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab27190, 1æg/ml) overnight at +4øC. The secondary antibody (green)ÿwas Alexa Fluor© 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor© 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43æM.

I am sorry to confirm that as far as we are aware, this antibody has not specifically been tested on brain samples in western blot so we are not able to provide this information from testing procedures. I can suggest that we usually recommend to load 20 - 30 ug of total protein lysate per lane of a gel in western blot. However, the exact amount may require some optimization for individual experiments. Reviewing the Unigene expression profile protein database link from the datasheet, this indicates there is medium level of expression in brain. Therefore, 20 -30 ug should be suitable for brain samples, but you can change this in order to optimize the results if necessary. I hope this will be helpful to you. I have been in touch with the source of this antibody and I can tell you that PDE10A (ab14622) was tested using rat, mouse and human PDE10A variants (PDE10A1 and DE10A2). However, based on sequence similarity the antibodies are expected to label the Chimpanzee, fowl and cat PDE10A proteins as well. I am sorry but I do not have an answer to the second part of your enquiry, an answer has not been forthcoming from the originator of this antibody. Thank you for your enquiry. I apologise for the delay in responding to your message. We have a high number of enquiries at the moment, and it has taken some time to obtain the information for you. The PDE10A antibody(ab14622) does not cross react with other PDE family members, however, it is known to label at least 2 variants of PDE10A. The cross reactivity against all the PDE4 isoforms has been checked, and all the PDE family representatives including PDD1 through to PDE11. There have been no crossreactivity studies done so far with the MGR5 antibody (ab27190), it may have very weak cross-reactivity with mGluR1,since the immunogen has 40%homology with mGluR1. However,WB results shows ab detects just a single band at about 130kDa(mGluR5 MW=130kDa,mGluR1 MW=142kDa). I hope this information is helpful. They also mentioned that the intensity of staining observed by this antibody relates to the tissue prepared. That is to say they recommend that an antibody titration experiment is performed.
