Properties

Applications

Our Abpromise guarantee covers the use of ab86233 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
IHC-P Use a concentration of 2.5 µg/ml.
WB Use a concentration of 1 µg/ml. Predicted molecular weight: 12 kDa. Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.

Target

  • Function

Associates with the organic matrix of bone and cartilage. Thought to act as an inhibitor of bone formation.
  • Involvement in disease

Defects in MGP are the cause of Keutel syndrome (KS) [MIM:245150]. KS is an autosomal recessive disorder characterized by abnormal cartilage calcification, peripheral pulmonary stenosis neural hearing loss and midfacial hypoplasia.
  • Sequence similarities

Belongs to the osteocalcin/matrix Gla protein family.
    Contains 1 Gla (gamma-carboxy-glutamate) domain.
  • Post-translational
    modifications

Requires vitamin K-dependent gamma-carboxylation for its function.
  • Cellular localization

    Secreted.

Answer

Thank you for contacting us.

The anti-MGP antibody (ab70193) has been raised against the full length MGP protein and as it is a polyclonal antibody the exact epitope cannot be determined. We would expect theantibodyto be able to detectboth forms of theprotein(yourmutated and unmutated constructs) howeverit is theoretically possible that the antibody is only able to detect your Thr variant. Unfortunately, there is no way of knowing this conclusively using this antibody and the only other antibody we have against this target (ab86233) has also been raised against an immunogen encompassing the residue 83.

Have you been able to see theAla mutantin your SDS? How is it beingexpressed?

I am sorry that I couldnot be of more help in this instance. If you have any further questions, please do not hesitate to contact us again.
