    Variant histone H2A which replaces conventional H2A in a subset of nucleosomes where it represses transcription. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling. May be involved in stable X chromosome inactivation.

Unfortunately, these kind of projects are not of a very high priority, especially as this additional band is clearly of a different size than the expected one.

According to ourBLAST searches there is no other protein that shares significant homologywith this region of MacroH2A2. Given that thisband is observedwith acell line derived from a tumour, it might be that it isdue to the specificbiology of this cancer cell line A431.

Please accept my apologies for the long delay of my reply. I have been in contact with the source of this antibody regarding the additional band between 72 and 55 kDa. We suspect that this band may be due to alternative splicing. Unfortunately, the identity of the protein causing this band has not been determined further. The lab however confirmed that when testing this antibody in mouse brain samples this band is only very faint.
