    Key regulator of mitochondrial calcium uptake required for calcium entry into mitochondrion. May act as a calcium sensor via its EF-hand domains, gating the activity of a calcium channel partner. Induces T-helper 1-mediated autoreactivity, which is accompanied by the release of IFNG.

Images

  • ICC/IF image of ab102830 stained HepG2 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab102830, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899, DyLight® 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Based on the information in your review, I feel that either one of the following could explain your results:
- The human protein has 5 isoforms, thus it might be possible that the rat protein also exists in several isoforms and you detected one of those.
- The other alternative is that the antibody does not detect the rat protein.

The immunogen sequence used to generate this antibody (ab102830) is excluded from this isoform, so this is not a possibility. Ab115025 would recognize this isoform, but itis not present on your blot.

Yes, this antibody is detecting Isoform 4 (MW 32kDa), as opposed to the other two which detected isoform 1 in the samples. You may still detect a 55kDa band in your samples if that is the isoform present.
