MM2 probe (ab172913) is a cell-penetrating phosphorescence-based O2 sensing probes enable direct high-resolution 2D and 3D mapping and quantitation of (intra)cellular O2. MM2 probe (ab172913) and NanO2 probe (ab172912), offer a simple and convenient approach to the direct real-time live-cell imaging in-vitro, in-vivo, and for 3D cultures on standard fluorescent microscopes (wide-field, confocal or two-photon).
Number of Assays: 10 - 100 assays
Number of tests vary depending on platform used in the assay. The probe contains sufficient material for either 100 assays in a standard 96 well plate or 10 assays in an imaging mini dish 35mm.
Probe Preparation and Cell Loading
Probe is supplied as 100 µg aliquots in plastic vials.
1. Add 100 µL of water to the vial and vortex for 30 seconds to produce 100X stock. Dilute stock with medium to working concentration 10 µg/mL.
2. Add this medium with probe to the cells, incubate at 37°C in CO2 incubator for 3-24h.

3. Wash the cells 1-2 times with fresh medium. Add medium, usually same volume as above.
4. Conduct optical measurements.
Ensure temperature equilibration of samples during the measurements since the probe is sensitive to both O2 and T.
