  • Function

    Has chymotrypsin-like activity. Hydrolyzes the amide bonds of synthetic substrates having Tyr and Phe residues at the P1 position. Preferentially hydrolyzes the 'Tyr-4-
    -Ile-5' bond of angiotensin I and the 'Phe-20-
    -Ala-21' bond of amyloid beta-protein, and is less active towards the 'Phe-8-
    -His-9' bond of angiotensin I and the 'Phe-4-
    -Ala-5' and 'Tyr-10-
    -Glu-11' bonds of amyloid beta-protein. Involved in thrombin regulation and fibronectin processing.

Please be aware that our ELISA assay is used for determining the activity of the antibody by applying serially diluted antibody to micro-wells that are coated with the immunizing peptide.

ELISA titre determination protcol:

Wells are coated with the immunizing peptide and serially diluted antibodies are applied and measured.

Coat 0.1ug peptide in 100ul PBS per well on high binding ELISA plates overnight at 4C.
Wash 3 times with 300ul PBS per well.
Block with PBS/0.1%(v/v) Tween-20 (PBST) containing 3%(w/v) skimmed milk (blocking buffer) for 1h at 37C
Wash 3 times with PBST
Make serially dilutions of antibody in blocking buffer and apply 100ul per well.
Incubate 1h at 37C with the plates covered
Wash three times with 300ul PBST
Apply secondary antibody (AP-conjugate) at appropriate dilution in blocking buffer, 100ul per well
Wash four times in 300ul PBST
Wash 2 times in 300ul PBS
Add 50ul per well p-nitro phenyl phosphate (pNPP) from Sigma and incubate for 30min at 30C
Stop the reaction with 50ul 1M NaOH.
Read at 405nm

Phosphate buffered saline (PBS): 20mM Sodium phosphate, pH7.4 in 150mM NaCl.

ab92368 is a Fast Track antibody. Fast-Track antibodies constitute a diverse group of products that have undergone variable degrees of characterization. They have all been affinity purified and show high titre values against the immunizing peptide (by ELISA). This antibody has been characterized initially in peptide ELISA i.e. the immunizing peptide was coated on the plate. The antibody detection limit is 1/64,000.

We do not have any ELISA data with other samples or even with natural endogenous protein; which is why we are selling this product at cheap price. Please read the full terms of use given on the datasheet.

This product is not covered under Abpromise guarantee because the return you will get from Abpoints will be approximately equal to the price you pay for this product.
