I want to measure the MMPs activation in mice lung fibrosis sample, and I found this kit from your company (ab112147 MMP Activity Assay Kit). I have two questions about this kit: 1. Can this kit use to measure multiple MMPs at one time? or only measure MMP3? 2. How many total protein that should load to each well?

Ab112147 is for total MMP activity assay, though we do offer MMP3 detection kits as well. This kit will not differentiate between the different MMPs. On the graph displayed on the datasheet distinct purified MMP enzymes were used. The graph shows the results achieved when using different forms of the MMP (bars 1-14 represent MMP1-MMP14 respectively). The detection limit of ab112147 is 1-10 ng/well of the MMPs, so it is really depends on how much MMPs in your sample, if it is 0.1%, 10ug/well should be enough. Use Tris buffer pH 7.5 for tissue homogenization.
