Properties

Applications

Our Abpromise guarantee covers the use of ab52897 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB 1/1000 - 1/10000. Detects a band of approximately 54 kDa (predicted molecular weight: 54 kDa).
IP 1/20.

For unpurified use at 1/30.

  • Application notes
Is unsuitable for Flow Cyt or IHC-P.
  • Target

    • Function

May be involved in tissue injury and remodeling. Has significant elastolytic activity. Can accept large and small amino acids at the P1' site, but has a preference for leucine. Aromatic or hydrophobic residues are preferred at the P1 site, with small hydrophobic residues (preferably alanine) occupying P3.
    • Tissue specificity

Found in alveolar macrophages but not in peripheral blood monocytes.
    • Sequence similarities

Belongs to the peptidase M10A family.
      Contains 4 hemopexin-like domains.
    • Domain

The conserved cysteine present in the cysteine-switch motif binds the catalytic zinc ion, thus inhibiting the enzyme. The dissociation of the cysteine from the zinc ion upon the activation-peptide release activates the enzyme.
    • Cellular localization

Secreted > extracellular space > extracellular matrix.
    • Information by UniProt
    • Database links

    • Form

The presence of a 22kDa truncated active form of MMP-12 in vitro as well as in vivo due to the internal autolytic cleavage of the C-terminus.

Diluting buffer and concentration: 5% NFDM /TBST.

    • ab52897 (purified) at 1/20 immunoprecipitating Glutamate Receptor 1 in HeLa whole cell lysate. 10 ug of cell lysate was present in the input. For western blotting, a HRP-conjugated Veriblot for IP Detection Reagent (ab131366) (1/1,500) was used for detection. A rabbit monoclonal IgG (ab172730) was used intead of ab128913 as a negative control (Lane 3).

Blocking buffer and concentration: 5% NFDM/TBST.

Diluting buffer and concentration: 5% NFDM /TBST.

    • Based on Western blot, unpurified ab52897 recognizes both pro and active MMP. The propeptide is 54 kDa, and 45 kDa and 22 kDa are active forms. The primary was used at 1:100 to detect recombinant MMP12 protein, but can be diluted more to decrease the intensity of the bands.

    Answer

I can confirm that both ab52897 and ab124964 are rabbit IgG monoclonals.

We have several suitable isotype controls available including the following. These have both been tested in IHC-P. I can recommend to review the online datasheets for further information.

    Answer

    Question
    Answer

Thank you for your enquiry. The antigen retrieval method used for this product was pressure cooker method. Please find the details of the protocol below. Pressure cooker antigen retrieval protocol. Solution: Tris-EDTA Buffer (10mM Tris Base, 1mM EDTA Solution, 0.05% Tween 20, pH 9.0) Tris 1.21 g EDTA 0.37 g Distilled water 1000 ml (100 ml to make 10x, 50 ml to make 20x) Mix to dissolve. pH is usually at 9.0. Add 0.5 ml of Tween 20 and mix well. Store at room temperature for 3 months or at 4C for longer storage. Protocol: 1. Immerse slides into staining dish containing Antigen Retrieval Solution. 2. Place covered staining dish into the rice cooker. Add 120 mL dH2O and press “cook”. 3. When “cook” is turned to “warm” (about 20–30 min), unplug the cooker and remove the staining dish to the bench top. 4. Allow to cool down, without cover, for 20 min.
