Target

  • Function

    Activator of LATS1/2 in the Hippo signaling pathway which plays a pivotal role in organ size control and tumor suppression by restricting proliferation and promoting apoptosis. The core of this pathway is composed of a kinase cascade wherein MST1/MST2, in complex with its regulatory protein SAV1, phosphorylates and activates LATS1/2 in complex with its regulatory protein MOB1, which in turn phosphorylates and inactivates YAP1 oncoprotein and WWTR1/TAZ. Phosphorylation of YAP1 by LATS1/2 inhibits its translocation into the nucleus to regulate cellular genes important for cell proliferation, cell death, and cell migration. Stimulates the kinase activity of STK38 and STK38L.

  • Paraffin-embedded human small intestine tissue stained for Mob1A using ab236969 at 1/100 dilution in immunohistochemical analysis.

  • HepG2 (human liver hepatocellular carcinoma cell line) cells stained for Mob1A (Green) using ab236969 at 1/333 dilution in ICC/IF, followed by Alexa Fluor 488-conjugated Goat Anti-Rabbit IgG (H+L) secondary antibody.

    The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. Counterstained with DAPI.

  • Mob1A was immunoprecipitated from 500 μg Jurkat (human T cell leukemia cell line from peripheral blood) whole cell lysate. For western blotting, a HRP-conjugated Protein G antibody was used as the secondary antibody (1/2000).

    Lane 1: Rabbit control IgG instead of ab236969 in Jurkat cell lysate.

    Lane 2: ab236969 IP in Jurkat cell lysate.

    Lane 3: Jurkat whole cell lysate (input).

  • Paraffin-embedded human breast cancer tissue stained for Mob1A using ab236969 at 1/1000 dilution in immunohistochemical analysis.

    After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum for 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
