    Apolipoprotein CIII in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of mouse Apolipoprotein CIII protein in serum, plasma, cell culture supernatant, and tissue extract samples.


    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.

  • Notes

    Apolipoprotein CIII is a surface component of chylomicrons, very low density lipoproteins (VLDL), and high density lipoproteins (HDL) in plasma. Intracellularly, Apolipoprotein CIII promotes hepatic very low density lipoprotein 1 (VLDL1) assembly and secretion. Extracellularly, Apolipoprotein CIII impairs the lipolysis of triglyceride-rich lipoproteins (TRLs) therefore decreasing clearance of TLRs. Mouse Apolipoprotein CIII is composed of a 20 amino acid signal peptide and a 79 amino acid mature chain. Human and rat Apolipoprotein CIII are 62% and 82% identical to mouse Apolipoprotein CIII, respectively.

Undiluted samples are as follows: serum 1:20,000, plasma (citrate) 1:20,000, plasma (EDTA) 1:20,000, and plasma (heparin) 1:20,000. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Apolipoprotein CIII concentration was determined to be 102.29 µg/mL in neat serum, 94.12 µg/mL in neat plasma (citrate), 111.95 µg/mL in neat plasma (EDTA), and 97.91 µg/mL in neat plasma (heparin).

  • The concentrations of Apolipoprotein CIII were measured in duplicate, interpolated from the Apolipoprotein CIII standard curves and corrected for sample dilution. Undiluted samples are as follows: lung supernatant 50% and liver supernatant 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Apolipoprotein CIII concentration was determined to be 13.17 ng/mL in neat lung supernatant and 36.64 ng/mL in neat liver supernatant.

The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
