    Mouse Bax ELISA Kit (ab233624) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Bax protein in tissue homogenate and cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse Bax with 19.4 pg/ml sensitivity.


    SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:


            - Single-wash protocol reduces assay time to 90 minutes or less
            - High sensitivity, specificity and reproducibility from superior antibodies
            - Fully validated in biological samples
            - 96-wells plate breakable into 12 x 8 wells strips


    A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

  • Function

    Accelerates programmed cell death by binding to, and antagonizing the apoptosis repressor BCL2 or its adenovirus homolog E1B 19k protein. Under stress conditions, undergoes a conformation change that causes translocation to the mitochondrion membrane, leading to the release of cytochrome c that then triggers apoptosis. Promotes activation of CASP3, and thereby apoptosis.
  • Tissue specificity

    Expressed in a wide variety of tissues. Isoform Psi is found in glial tumors. Isoform Alpha is expressed in spleen, breast, ovary, testis, colon and brain, and at low levels in skin and lung. Isoform Sigma is expressed in spleen, breast, ovary, testis, lung, colon, brain and at low levels in skin. Isoform Alpha and isoform Sigma are expressed in pro-myelocytic leukemia, histiocytic lymphoma, Burkitt's lymphoma, T-cell lymphoma, lymphoblastic leukemia, breast adenocarcinoma, ovary adenocarcinoma, prostate carcinoma, prostate adenocarcinoma, lung carcinoma, epidermoid carcinoma, small cell lung carcinoma and colon adenocarcinoma cell lines.

      The Bax standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

      Interpolated concentrations of native Bax in mouse C2C12 cell extract based on a 12.5 µg/mL extract load, A20 cell extract based on a 5 µg/mL extract load, and PC-12 cell extract based on a 25 µg/mL extract load. The concentrations of Bax were measured in duplicate and interpolated from the Bax standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Bax concentration was determined to be 3,001 pg/mL in C2C12 cell extract, 1,404 pg/mL in A20 cell extract, and 1,689 pg/mL in PC-12 cell extract.

      Cells were collected directly after treatment and subcellular fractions were prepared using a cell fractionation kit (ab109719). The concentrations of Bax were measured in three different dilutions of the fraction samples in duplicate and interpolated from the Bax standard curve. The interpolated values are plotted (mean +/- SD, n=3). The mean Bax concentration was determined to be 841.4 pg/mL in the untreated cytosol fraction, 100.0 pg/mL in the untreated mitochondrial fraction, 912.6 pg/mL in the untreated whole cell sample, 186.9 pg/mL in the treated cytosol fraction, 1,366 pg/mL in the treated mitochondrial fraction, and 1,622 pg/mL in the treated whole cell sample.
