  • Product overview

Abcam’s Fetuin A (AHSG) in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Fetuin A protein in mouse serum, plasmas, cell culture supernatants and tissue extracts.

The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.

    Sensitivity:

Samples diluted in Sample Diluent NS + Enhancer: 0.682 ng/mL

Samples diluted in 1X Cell Extraction Enhancer: 0.312 ng/mL

  • Notes

Fetuin A is a highly abundant protein secreted by the liver that is capable of binding a wide variety of ligands, similar to serum albumin. A member of the cystatin family, Fetuin A plays a vital role in preventing tissue calcification and is also involved in fatty acid transport, insulin regulation and anti-inflammatory responses. Fetuin A is encoded by a single gene on chromosome 3, contains two cystatin domains, and exists as a heterodimer in its posttranslational form. Fetuin A is more abundant than serum albumin in fetal serum.

  • SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

  • Standard curve comparison between mouse Fetuin A SimpleStep ELISA® kit and traditional ELISA kit from leading competitor. SimpleStep ELISA kit shows comparable sensitivity with shorter protocol time.

  • Background-subtracted data values (mean +/- SD) are graphed.

  • The 1X dilution was 0.002% for the mouse serum and 0.003% for all of the mouse plasmas. Interpolated data values (mean +/- SD, n = 2) are graphed.

  • A blank cell culture media control was also analyzed, and was negative. The 1X dilution was 10.0% for the unstimulated liver supernatant and 12.5% for the unstimulated spleen supernatant. Interpolated data values (mean +/- SD, n = 2) are graphed.

  • The 1X dilution was 250 µg/mL for the liver extract and 62.5 µg/mL for the lung extract. Interpolated data values (mean +/- SD, n = 2) are graphed.
