    Abcam’s Fractalkine (CX3CL1) in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Fractalkine protein in mouse serum, plasma and cell culture supernatants.

    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.

  • Notes

    Fractalkine (CX3CL1 or Neurotactin) is a chemokine produced from a type I transmembrane protein on the cell surface. The soluble chemokine is generated by proteolytic cleavage and represents residues 25-100 of the full protein sequence. Fractalkine expression is primarily on epithelial cells. Fractalkine functions both in the full length transmembrane form (for adhesion) and in the released soluble form (chemoattraction).

  • Relevance

    CX3CL1 is a member of the delta chemokine subfamily that contains a unique CX3C cysteine motif near the N-terminal. Unlike other known chemokines, it is a type 1 membrane protein containing a chemokine domain tethered on a long mucin-like stalk. CX3CL1, a leukocyte chemoattractant, is expressed in various tissues including the brain, heart, lung, kidney, skeletal muscle, and testis. The expression is reported to be up-regulated in endothelial cells and microglia by inflammatory signals. CX3CR1, a specific receptor for fractalkine, mediates both leukocyte migration and adhesion.

  • The concentrations of Fractalkine were measured in duplicate and interpolated from the Fractalkine standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Fractalkine concentration was determined to be 354 pg/mL in mouse serum, 328 pg/mL in mouse plasma (Heparin), 350 pg/mL in mouse plasma (EDTA), and 2152 pg/mL in L929 stimulated cell culture media.

  • L929 cells were grown in the absence (unstimulated) or presence of Phorbol Myristate Acetate (PMA) and phytohemagglutinin (PHA) (stimulated) for 3 days. Fractalkine was measured in 2-fold diluted cell culture supernatants of unstimulated and PMA/PHA stimulated L929 and cell culture media. Measured values were interpolated from the Fractalkine Standard Curve diluted in Sample Diluent NS and corrected for dilution factor. Mean of duplicate values +/-SD are graphed: 704 pg/mL unstimulated, 2136 pg/mL stimulated, and undetectable in media.

Assay was performed using the Abcam Kit protocol. Blood samples were obtained from 15-17 week-old male C57Bl/6 mice and serum samples were prepared and stored as recommended in the Abcam Kit protocol. Serum samples were analyzed diluted 1:2. All sample concentrations were within the kit detection range. CX3CL1 serum level in healthy control group was 4068±360 pg/mL.
