    Abcam’s Galectin 3 Mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse Galectin 3 in serum, plasma and cell culture supernatants.

    This assay employs an antibody specific for Mouse Galectin 3 coated on a 96- well plate. Standards and samples are pipetted into the wells and Galectin 3 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse Galectin 3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Galectin 3 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

    Galactose-specific lectin which binds IgE. May mediate with the alpha-3, beta-1 integrin the stimulation by CSPG4 of endothelial cells migration. Together with DMBT1, required for terminal differentiation of columnar epithelial cells during early embryogenesis By similarity. In the nucleus: acts as a pre-mRNA splicing factor. Involved in acute inflammatory responses including neutrophil activation and adhesion, chemoattraction of monocytes macrophages, opsonization of apoptotic neutrophils, and activation of mast cells. Probably forms homo- or heterodimers. Interacts with DMBT1. Forms a complex with the ITGA3, ITGB1 and CSPG4. Interacts with LGALS3BP, LYPD3, CYHR1 and UACA. A major expression is found in the colonic epithelium. It is also abundant in the activated macrophages. Contains 1 galectin domain.

    I am using tissue homogenate as my sample for use with this kit. Maybe homogenate has not been tested before but there is no inidation as to which Diluent to use (A or B) with this type of sample.
    1. What is the difference between diluent A and B?
    2. Which diluent do you recommend for use with tissue homogenate?

    Answer

    While this ELISA kit was neither designed nor validated with tissue lysate samples, our kits are very accommodating to many different samples types so lysates will most likely work. If you decide to test these kits with tissue lysate samples, we recommend diluting the samples at least 5-fold with Assay Diluent B to minimize any effects of the detergents in the lysis buffer. The samples may need to be diluted further but this would need to be determined by the experimenter empirically.

    Concentration of the original lysate should ideally be at least 1 mg/ml protein, preferably more, to achieve 50-500 ug/ml after dilution. Please note this range should just be used as a starting point and the optimal concentration would need to be determined empirically.


    In general, we recommend your lysis buffer meet the following specifications:
    A) has relatively low salt content (700 mM or less)
    B) does not contain sodium azide
    C) does not contain >0.1% SDS
    D) does not contain >10 mM reducing agents (beta-mercaptoethanol or dithiothreitol)
