    Abcam’s Galectin-7 in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of mouse Galectin-7 protein in serum, plasma, and tissue extracts.

    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.

    Galectin-7 is a 15kDa monomeric member of the galectin family of carbohydrate-binding proteins. Galectin-7 is primarily expressed in epithelial cells. It is hypothesized to have roles in cell-cell and/or cell-matrix interactions necessary for normal growth control.

    Galectins belong to a family of related beta-galactoside-binding lectins, also referred to as S-type or S-Lac lectins. Members of this family have been implicated in a variety of functions, including growth regulation, cell adhesion, migration, neoplastic transformation, and immune responses. Galectin 7 is thought to be involved in cell-cell and/or cell-matrix interactions necessary for normal growth control. Galectin 7 has been implicated in apoptosis and is believed to upregulate MMP9 and as such be involved in the progression of tumorigenesis.

The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Galectin-7 concentration was determined to be 5.07 ng/mL in neat mouse serum, 3.56 ng/mL in neat mouse plasma (EDTA), and 4.36 ng/mL in neat mouse plasma (Heparin).

The mean galectin-7 concentration was determined to be 2.06 ng/mL in 2 µg/mL mouse skin extract and 1.39 ng/mL in 5 µg/mL mouse esophagus extract.
