Mouse heart tissue lysate - mitochondrial extract (ab110350)

Overview

  • Product name
Mouse heart tissue lysate - mitochondrial extract
    See all Mouse Heart lysates
  • General notes

Product was previously marketed under the MitoSciences sub-brand.

2 mg of mouse heart mitochondria in 400ul of iso-osmotic buffer. This sample can act as a positive control in immunocapture applications and Blue Native PAGE.
1 mg solubilized mitochondria for Immunocapture experiments. For the Immunocapture of OXPHOS complexes it is recommended that 180 µl of the mitochondrial membranes supplied be solubilized by the addition of 20 µl of 10% lauryl maltoside. Insoluble material is removed by centrifugation at 72,000g before immunocapture of complexes from the supernatant.

Answer

Thank you for contacting us.

This product description is somewhat misleading. It is a mitochondrial fraction from pooled mouse hearts: hearts are minced and outer membranes ruptured by dounce homogenization, mitochondria are isolated by differential centrifugation. Mitochondria are provided in an iso-osmotic buffer (on PWS). This is not a lysate. We can make no claim as to how intact the mitochondria are following isolation and freeze/thaw, but we know inner and outer membranes and matrix are present and suitable for immunoprecipitations or SDS-PAGE. Also, this is an enriched mitochondrial fraction – it is not fully purified – we expect other cellular membranes to be present (e.g. some contribution of plasma membrane, other organelle, very minor contribution of nucleus)

1/3. This preparation is from a pool of 138 adult mouse hearts, mixed strains. We do not have record of strains of mice contributing hearts.

2. Mitochondria

3. For mitochondrial DNA/RNA – we have not tried ourselves. For DNA: yes, should work - mtDNA nucleoids will be present (possibly also some nuclear DNA). For RNA: use caution – no steps during mitochondrial isolation taken to stabilize RNA.
