How to do 1/1000 dilution without losing too much assay buffer?

Regarding the dilution, there is really no way to do a 1000-fold dilution and use < 30 ml of diluent testing 40 samples without doing a 2-step dilution. I’d give the below example of a 2-step dilution to conserve both sample and diluent:
1.2ul of sample + 18ul of diluent = 20ul of 10-fold diluted sample
2.2ul of 10-fold diluted sample + 198ul of diluent = 200ul of 1000-fold diluted sample (enough for duplicate wells)
Please follow these steps, the solution will be more than enough.
