    IL-4 in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of IL-4 protein in mouse serum, cell culture supernatant, and cell and tissue extract samples.


    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.

  • Notes

    Mouse IL-4 is a 120 amino acids long secreted protein. IL-4 is a cytokine produced mostly by activated T lymphocytes, mast cells and basophils. It has variety of immune response modulating functions by acting on number of cell types. IL-4 participates in several B-cell activation processes. It is a co-stimulator of DNA-synthesis. It induces the expression of class II MHC molecules on resting B-cells. It enhances both secretion and cell surface expression of IgE and IgG1. It also regulates the expression of the low affinity Fc receptor for IgE (CD23) on both lymphocytes and monocytes. Positively regulates IL31RA expression in macrophages.

    Genetic variations in IL4 may be a cause of susceptibility to ischemic stroke (ISCHSTR) [MIM:601367]; also known as cerebrovascular accident or cerebral infarction. A stroke is an acute neurologic event leading to death of neural tissue of the brain and resulting in loss of motor, sensory and/or cognitive function. Ischemic strokes, resulting from vascular occlusion, is considered to be a highly complex disease consisting of a group of heterogeneous disorders with multiple genetic and environmental risk factors.

  • Comparison of IL-4 in unstimulated and PHA+PMA stimulated EL4.IL2 cell supernatants. EL4.IL2 cells were cultured for 2 days in the presence or absence of 10 µg/mL PHA and 10 ng/mL PMA. The concentrations of IL-4 were measured in three different dilutions of the supernatant samples in duplicates and interpolated from the IL-4 standard curve. The interpolated values are plotted (mean +/- SD, n=3). The mean IL-4 concentration was determined to be 3549 pg/mL in PHA+PMA stimulated EL4.IL2 cell culture supernatant and undetectable in unstimulated EL4.IL2 cell culture supernatant and DMEM media (not shown).

  • Interpolated concentrations of native IL-4 in PHA+PMA treated EL4.IL2 cell extract samples based on a 500 μg/mL extract load. The concentrations of (target) were measured in duplicate and interpolated from the IL-4 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-4 concentration was determined to be 822.0 pg/mL in PHA+PMA treated cell extract samples and undetectable in untreated EL4.IL2 cell extract samples (results not shown).

  • Interpolated concentrations of native IL-4 in PHA+PMA treated EL4.IL2 cell culture supernatant (4 days) samples. The concentrations of IL-4 were measured in duplicates, interpolated from the IL-4 standard curves and corrected for sample dilution. Undiluted samples are as follows: EL4.IL2 cell supernatant 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-4 concentration was determined to be 3529 pg/mL in neat PHA+PMA treated EL4.IL2 cell culture supernatant.

  • Interpolated concentrations of spike IL-4 in mouse serum samples.
