    IP-10 (CXCL10) in vitro SimpleStep ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of IP-10 protein in mouse serum, plasma and cell culture supernatants.


    The SimpleStep ELISA employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.

  • Notes

    C-X-C motif chemokine 10 (CXCL10 or IP-10) is a small 10.8kD protein that is secreted by several cell types in response to interferon-gamma (IFNg). These cell types include monocytes, endothelial cells and fibroblasts. Upon secretion, CXCL10 is cleaved into an 8.7kD biologically active protein to function in chemotaxis for T-cells, NK cells, monocytes/macrophages and dendritic cells. In addition, CXCL10 has antitumor activity through the inhibition of bone marrow colony formation and angiogenesis. CXCL10 elicits its effects by binding to the cell surface chemokine receptor 3 (CXCR3).

  • The concentrations of IP-10 were measured in duplicates, interpolated from the IP-10 standard curves and corrected for sample dilution. Undiluted samples are as follows: 4% supernatant. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IP-10 concentration was determined to be 21.7 g/mL in in RAW 264.7 supernatant.

  • Native IP-10 was measured in duplicate in 50% serum and 50% EDTA plasma diluted in Sample Diluent 25BP and concentrations interpolated from a standard curve diluted in Sample Diluent 25BP. The interpolated dilution factor corrected values are graphed (mean +/- SD).
